Welch Postdoctoral Research Associate : Performing the s ynthesis of the modified RNAs and studies of their structures, dynamics, and interactions with the proteins. Current projects:
- Structural studies of the anticodon stem loop (ACSL) of tRNA Phe containing pseudouridines and N6-dimethylallyl-modified adenosine. These require the preparation of 13C, 15N-labeled RNAs using enzymatic methods, the expression and purification of the unlabeled and 13C, 15N-labeled proteins in the E. coli bacteria, the chemical synthesis of site 13C, 15N-specific labeled nucleotides and the application of the 2D and 3D NMR techniques to solve the structural problems.
- Determination the metal binding sites in the complex thio-modified rRNA (helix21 of 16S rRNA) with the ribosomal S8 protein from E. coli using NMR and EPR techniques.
- Chemical synthesis of site specific 15N-labeled nucleosides used to find the mechanism reaction of the RNA with the MiaA enzyme (tRNA isopentenyl transferase).
- Structural studies of the small nucleolar RNA (snoRNA) H/ACA box involved in rRNA pseudouridylation in Euglena gracillis using NMR and ITC techniques.
