Adenylate kinases are small enzymes that catalyze the interconversion of adenylate species (AMP, ADP, ATP), maintaining their homeostasis in the cell. AK’s from a variety of organisms provide a unique opportunity to study relations between functions, dynamics and protein stabilities. There are two oligomeric classes of AK: monomeric as found in Eubacteria and trimeric in Archaebacteria. I am studying monomeric Marinibacillus marinus AK, and trimeric AK from Methanococcus maripaludis, a mesophilic methanogenic Archaebacteria using a combination of; X-ray crystallography, circular dichroism and enzyme activity assays to gain a better understanding of the evolution of protein thermostability and activity differences in the AK family of enzymes.
